2020 Optimized Serial Expansion of Human Induced Pluripotent Stem Cells Using Low-Density Inoculation to Generate Clinically Relevant Quantities in Vertical-Wheel Bioreactors

Abstract

Describes optimization of low-density inoculation, oxygen, agitation, and feeding strategies for serial iPSC expansion in Vertical-Wheel® bioreactors. The process achieved more than 30-fold expansion in 6 days and over one million-fold cumulative expansion across serial passages while maintaining pluripotency, normal karyotype, and teratoma-forming potential.
